An integrative dynamic systems approach to understanding Src family kinase signalling
This Ph.D. project focussed on protein interactions involved in the regulation of the tyrosine kinase Src, which plays a major role in a number of signal transduction pathways. From the multitude of known protein interactions described in the literature, a differential equations model was defined and analysed. The simulations provided insights into systems-level properties of the network, namely a robust, bistable switch and excitable behaviour. A Monte Carlo investigation of the parameter space was used to quantify the robustness of the network and to assess the likelihoods of two alternative models, representing mechanistic hypotheses inspired from literature. A further study addressed issues relating to the interface between Src family kinases and several receptor tyrosine kinases. The model serves as a coherent and quantifiable description of Src family kinase signalling events and makes novel, experimentally testable predictions.
