The ensuing years (2005-2018) generated ample confirmatory observations, which are summarized in his subsequent papers. The mechanism proposed is accrual of somatic mutations in individual neurons of the behavioral-motor neural super-network. Accrual of a critical combination of changes in a single neuron generates a cascade of destructive changes that spread through the entire network. These changes lead to accelerated neuronal loss and systematic dismantling of the network. Biological techniques now available have confirmed the accrual of somatic mutations in individual cortical neurons, and their relation to neurodegenerative diseases. An additional article explores the ethical challenges of conducting clinical trials in patients with ALS.< Less
