A son arrivée en Amérique, la famille s’installe en Pennsylvanie, Etat fondé par les Quakers. Le jeune Antoine, à son tour, rejoint ce groupe et abandonne rapidement sa carrière dans le négoce pour devenir éducateur et professeur dans une école quaker tout en donnant chez lui des cours du soir pour des enfants d’esclaves. En 1754, il fonde la première école de filles d’Amérique, et quelques années plus tard il fonde une école pour les Africains-américains. et se bat contre contre l'esclavage.
